•In this study, 48
subjects (82.76%) were detected as fertile. Among the fertile subjects, 26
subjects (54.16%) did not showed any growth, 6 subjects (12.5%) were detected
for Ureaplasma
urealyticum , and 16 subjects
(33.33%) were detected for other organisms. Fried egg colony was detected in
blood agar after 72 hrs (without Co2) in 10 samples. We
inoculated the fried egg colony in the A7 agar, and broth in the presence of CO2 (using
candle jar technique). The A7 broth turns to yellow within 48 hrs of
inoculation. We also detected fried egg colony within 48 hrs in the A7 agar
medium. Colonies were found in the Zone of inhibition was detected for azithromycin and doxycyclin.
•Infections of the
male genitourinary tract account for up to 15% of cases of male infertility (Pellati D,). Acute and
chronic infections and consequent inflammation in the male reproductive system
may compromise the sperm cell function and the whole spermatogenetic process,
causing qualitative and quantitative sperm alterations (Henkel R, Sanocka-Maciejewska D). Recent studies
have shown that the simple presence of bacteria in semen samples may compromise
the sperm quality. The bacteria responsible for semen contaminations generally
originate from the urinary tract of patients or can be transmitted by the partner
via sexual intercourse
SUMMARY
•Semen sample was
collected from 58 male subjects (mean age = 33), who were referred to Kanmani Fertilty Clinic, Chennai,
India. A consent form was taken from the subjects before the sample
collection. Sterile sample was collected
for microbiological investigation. Semen analysis was done based on WHO
criteria. All the semen parameters were noted. The semen sample was inoculated
in Blood agar, Mac Conkey agar and A7 agar.
Blood agar and A7 agar showed discrete fried egg colonies of Ureaplasma
urealyticum. A7 agar is the
selective media for Ureaplasma urealyticum, still for further
confirmation Biochemical analysis is done. Negative results were observed for
all the IMViC reaction and
positive result was observed in urease test. Antimicrobial sensitivity test reveals Azithromycin and doxycycline has effect on Ureaplasma
urealyticum.
Ureaplasma urealyticum showed enhanced
growth in agar plate with Omega - 3 when compared with the normal A7 agar
plate. Further studies are required to determine the mechanism behind the
enhanced growth while addition of Omega – 3. In future PCR identification of
the Ureaplasma
urealyticum
will be done.
